I have set up my sister business with UK2 but I'm unable to send email
via Orange, I have testing on my own personal BT account and it works fine, so I know the problem is with Orange.

Orange don't allow external connections on port 25, default for sending email.
There are 2 real options:
1. Use the Orange SMTP server as the message suggests in order to send email.
2. Use a different server on an alternative port other than 25, the email server must allow connections on a different port to do this so you'd need to check with UK2 to see if they do.

"Can someone please advise how to solve the following problem" - yep you need to do exactly what the server has suggested in the error report: change the outgoing SMTP server set in your mail proogramme back to Orange and not via uk2.
Orange capture any Port 25 traffic (Port 25 being the default SMTP port) so when its set to something other than smtp.orangewhatever you will get their 554 error.
You could check if the UK2 server will accept smtp connections on a port other than 25 - if so you should be able to make use of that as it will bypass the Orange "Filters".
Many, not all, but many ISPs including Orange block outgoing port 25, its an anti spam measure. It stops you getting an email from your ISP complaining your trojan infected PC is spewing spam to all and sundry. As I got from Zen last week.... (School network, caretakers home machine infected with c**p).
